Regio-, Diastereo-, and Enantioselective Synthesis of Vicinal Diols via alpha-Silyl Ketones

A new versatile and efficient regio-, diastereo-, and enantioselective synthesis of vicinal diols s-trans-4, s-trans-5, and s-cis-4 is described.Symmetrical ketones are converted into their SAMP- or RAMP-hydrazones which are then silylated with (isopropyloxy)dimethylsilyl chloride, followed by ozonolysis to afford the alpha-silyl ketones (R)-2 of high enantiomeric purity (ee 90 > 98percent).On the other hand, methyl ketones, after conversion into the corresponding (-)-(S)-1-amino-2-(methoxymethyl)pyrrolidine (SAMP) hydrazones, are silylated and then alkylated with R1 to afford unsymmetrical alpha-silyl ketones (S)-3 of high enantiomeric purity (ee 90->98percent).The reduction of the above obtained alpha-silyl ketones with L-Selectride, followed by oxidative cleavage of the C-Si bond gives rise to s-trans-4, s-trans-5, and s-cis-4 with high diastereoselectivity (de 95->98percent) and without racemization (ee >90->98percent).

Efficient (3S)-acetoin and (2S, 3S)-2, 3-butanediol production from meso-2, 3-butanediol using whole-cell biocatalysis

(3S)-Acetoin and (2S, 3S)-2, 3-butanediol are important platform chemicals widely applied in the asymmetric synthesis of valuable chiral chemicals. However, their production by fermentative methods is difficult to perform. This study aimed to develop a whole-cell biocatalysis strategy for the production of (3S)-acetoin and (2S, 3S)-2, 3-butanediol from meso-2, 3-butanediol. First, E. coli co-expressing (2R, 3R)-2, 3-butanediol dehydrogenase, NADH oxidase and Vitreoscilla hemoglobin was developed for (3S)-acetoin production from meso-2, 3-butanediol. Maximum (3S)-acetoin concentration of 72.38 g/L with the stereoisomeric purity of 94.65% was achieved at 24 h under optimal conditions. Subsequently, we developed another biocatalyst co-expressing (2S, 3S)-2, 3-butanediol dehydrogenase and formate dehydrogenase for (2S, 3S)-2, 3-butanediol production from (3S)-acetoin. Synchronous catalysis together with two biocatalysts afforded 38.41 g/L of (2S, 3S)-butanediol with stereoisomeric purity of 98.03% from 40 g/L meso-2, 3-butanediol. These results exhibited the potential for (3S)-acetoin and (2S, 3S)-butanediol production from meso-2, 3-butanediol as a substrate via whole-cell biocatalysis.

Wavelength and mass resolved resonance-enhanced multiphoton ionization (REMPI) excitation spectra of (R)-(+)- 1-phenyl-1-propanol (PR) and its complexes with some chiral diols, i.e. 1,2-propanediols. 2.3-butanediols, and 2,4-pentanediols, have been recorded after a supersonic molecular beam expansion and interpreted in the light of molecular dynamic (MD) conformational minima searches. The spectral features of the selected complexes were found to depend on cooperative hydrogen-bond interactions between the two components, whose intensity depends upon the specific configuration of the diol moiety and the relative position of its hydroxy groups. The study further confirms resonant two-photon ionization spectroscopy, coupled with time-of-flight mass resolution (R2PI-TOF), as an excellent tool for gathering valuable information on the interactive forces in molecular clusters and for enantiodiscrimination of chiral molecules in the gas phase.

Biocatalytic production of alpha-hydroxy ketones and vicinal diols by yeast and human aldo-keto reductases

The alpha-hydroxy ketones are used as building blocks for compounds of pharmaceutical interest (such as antidepressants, HIV-protease inhibitors and antitumorals). They can be obtained by the action of enzymes or whole cells on selected substrates, such as diketones. We have studied the enantiospecificities of several fungal (AKR3C1, AKR5F and AKR5G) and human (AKR1B1 and AKR1B10) aldo-keto reductases in the production of alpha-hydroxy ketones and diols from vicinal diketones. The reactions have been carried out with pure enzymes and with an NADPH-regenerating system consisting of glucose-6-phosphate and glucose-6-phosphate dehydrogenase. To ascertain the regio and stereoselectivity of the reduction reactions catalyzed by the AKRs, we have separated and characterized the reaction products by means of a gas chromatograph equipped with a chiral column and coupled to a mass spectrometer as a detector. According to the regioselectivity and stereoselectivity, the AKRs studied can be divided in two groups: one of them showed preference for the reduction of the proximal keto group, resulting in the S-enantiomer of the corresponding alpha-hydroxy ketones. The other group favored the reduction of the distal keto group and yielded the corresponding R-enantiomer. Three of the AKRs used (AKR1B1, AKR1B10 and AKR3C1) could produce 2,3-butanediol from acetoin. We have explored the structure/function relationships in the reactivity between several yeast and human AKRs and various diketones and acetoin. In addition, we have demonstrated the utility of these AKRs in the synthesis of selected alpha-hydroxy ketones and diols.

Physicochemical characterization and quality of Dangshan pear wines fermented with different Saccharomyces cerevisiae

Three commercial yeasts strains, namely, Saccharomyces cerevisiae SY, DV10, and Drop Acid Yeast, were used for Dangshan pear wine fermentation. Monitoring main physical and chemical indexes and scoring comprehensive sensory characteristics to find a suitable yeast to produce Dangshan pear wine. The fermentation cycle of SY was short (15?days), and the SY-fermented wine had a suitable sugar-acid ratio, with a residual sugar content of 3.13?¡À?0.05?g/L, total acid content of 3.40?¡À?0.11?g/L, and ethanol content of 14.1?¡À?0.27% (v/v). Additionally, 42 flavor compounds were detected in fermented Dangshan pear wine, and the total amount of flavor compounds was highest in the SY wine (2,584.72?mug/L). Combined with the comprehensive sensory evaluation scores, these results suggest that Saccharomyces cerevisiae SY was the most suitable strain to produce Dangshan pear wine. Practical applications: In this study, we compared the physical and chemical indicators of pear wine brewed by different Saccharomyces cerevisiae in the process of fermentation and the final quality of pear wine products. It was concluded that the pear wine produced by Saccharomyces cerevisiae SY had good quality. The study found a strain suitable for the fermentation of pear wine and provided a theoretical basis for the industrial production of pear wine. Next, we can try to use Saccharomyces cerevisiae SY for large-scale production of pear wine and try to sell it on the market.
