The use of MAZP for the regeneration of the cofactors was investigated by applying the electrochemical oxidation of the mediator. The total turnover numbers of mediator MAZP and cofactor NADH for ethanol oxidation catalyzed by NAD+-dependent alcohol dehydrogenase depended on the concentration of the substrate and the duration of the electrolysis, and the yield of the reaction was limited by the enzyme inactivation and the electrochemical process. The PQQ-dependent alcohol dehydrogenase was more stable, and the turnover number of the enzyme reached a value of 2.3 × 103. In addition, oxidation of 1,2-propanediol catalyzed by the PQQ-dependent alcohol dehydrogenase proceeded enantioselectively to yield L-lactic acid.

The alpha-hydroxy ketones are used as building blocks for compounds of pharmaceutical interest (such as antidepressants, HIV-protease inhibitors and antitumorals). They can be obtained by the action of enzymes or whole cells on selected substrates, such as diketones. We have studied the enantiospecificities of several fungal (AKR3C1, AKR5F and AKR5G) and human (AKR1B1 and AKR1B10) aldo-keto reductases in the production of alpha-hydroxy ketones and diols from vicinal diketones. The reactions have been carried out with pure enzymes and with an NADPH-regenerating system consisting of glucose-6-phosphate and glucose-6-phosphate dehydrogenase. To ascertain the regio and stereoselectivity of the reduction reactions catalyzed by the AKRs, we have separated and characterized the reaction products by means of a gas chromatograph equipped with a chiral column and coupled to a mass spectrometer as a detector. According to the regioselectivity and stereoselectivity, the AKRs studied can be divided in two groups: one of them showed preference for the reduction of the proximal keto group, resulting in the S-enantiomer of the corresponding alpha-hydroxy ketones. The other group favored the reduction of the distal keto group and yielded the corresponding R-enantiomer. Three of the AKRs used (AKR1B1, AKR1B10 and AKR3C1) could produce 2,3-butanediol from acetoin. We have explored the structure/function relationships in the reactivity between several yeast and human AKRs and various diketones and acetoin. In addition, we have demonstrated the utility of these AKRs in the synthesis of selected alpha-hydroxy ketones and diols.

Complexes [Pd(eta3-CH3-C3H5l)P2]X (X = PF6, 4f, 4g; and X = BPh4, 5f), where P are monodentate diamidophosphite ligands with diazaphospholidine heterocyclic backbone obtained from (R,R)- and (S,S)-N,N?-dibenzylcyclohexane-1,2-diamine and alcoxy groups coming from (R)-phenyl-ethanol and (S)-borneol have been also prepared. Neutral palladium complexes [PdCl2(P-P)] (1a, 1c) were synthesized to prove the C2symmetry of the P-P ligand. The new compounds were fully characterized in solution by NMR spectroscopy. The X-ray crystal structure determination for 2e-(R,R,Ral,Ral;R,R) and 1a-(S,S;Sal,Sal;S,S) has been achieved. The new allyl-palladium complexes were applied in the asymmetric allylic substitution reaction of the benchmark substrate rac-3-acetoxy-1,3-diphenyl-1-propene with dimethyl malonate and benzylamine as nucleophiles in order to test their catalytic potential. The best results were obtained with the 3a-(R,R;Ral,Ral;R,R) precursor (up to 84% ee) while complexes with the e ligand derived from the (R,R)-N,N?-dimethylcyclohexane-1,2-diamine terminal fragment resulted inactive in the process. The influence of the nature and the absolute configuration of both the bridging and the terminal fragments of the bis(diamidophosphite) ligand on the asymmetric induction is discussed. A preliminary study of the anion effect (PF6?vs. BPh4-) on the activity and the enantioselectivity of the Pd-catalysed allylic substitution has also been performed.
